    摘要:目的 研究补肾健脾、益精填髓法对骨质疏松症模型大鼠骨代谢、骨生物力学的影响。方法 选用清洁级Wistar大鼠,按照随机分组方法60只大鼠分为手术组(48只)和正常组(12只)。将大鼠双侧卵巢摘除以制备骨质疏松症模型。术后7d随机将手术组大鼠分为4组,每组12只,即假手术组、模型组、右归丸组和实验组。干预12周,采集24 h内尿液后,处死大鼠取主动脉血及左侧股骨,进行代谢指标及骨生物力学测试。结果 (1)各组大鼠血Ca、P浓度比较,差异无统计学意义(P>0.05)。模型组ALP水平显著升高,与正常组及假手术组比较,差异有统计学意义(P<0.001)。右归丸组ALP水平与正常组及假手术组比较,差异无统计学意义(P>0.05)。实验组ALP水平显著低于其他组,差异均有统计学意义(P<0.001)。(2)模型组尿Ca/Cr、尿P/Cr显著高于正常组、假手术组(P<0.001)。右归丸组Ca/Cr、P/Cr与正常组、假手术组比较,差异无统计学意义(P>0.05)。实验组显著Ca/Cr、P/Cr显著低于其他组(P<0.05)。(3)模型组PINP、BMP-2低于其他各组,差异有统计学意义(P<0.001)。实验组PINP、BMP-2浓度高于右归丸组和模型组(P<0.05),而与正常组和假手术组无显著差异(P>0.05)。(4)模型组骨密度显著低于其他组别,差异有统计学意义(P<0.001)。实验组骨密度显著高于模型组、右归丸组(P>0.05),而与正常组、假手术组无显著差异(P>0.05)。模型组的骨强度显著低于其他组别,差异有统计学意义(P<0.001)。结论 补肾健脾、益精填髓法具有对抗骨质疏松的效果,且作用机制可能与其调整肠道微生态环境,减少骨矿物质丢失,上调骨组织中BMP-2表达,增加成骨细胞活性有关。

    Abstract:

    Abstract: Objective To investigate the effects of the methods of effects of tonifying kidney and spleen, benefiting essence and filling marrow on bone metabolism and bone biomechanics in osteoporosis model rats. Methods Wistar rats of clean grade were selected, and 60 rats were divided into operation group (48 rats) and normal group (12 rats) according to the random grouping method. The rat's bilateral ovaries were removed to prepare an osteoporosis model. 7 days after the operation, the surviving rats in the operation group were randomly divided into 4 groups, each with 12 rats: Sham operation group, model group, Yougui Pill group and experimental group. After 12 weeks of intervention, the rats were killed after collecting urine within 24 hours. Aortic blood and left femur were taken for metabolic indexes and bone biomechanics tests. Results (1) There was no significant difference in the concentrations of serum Ca and P in each group (P > 0.05). The level of (alkaline phosphatase,ALP)ALP in the model group was significantly higher than that in the normal group and the sham operation group (P < 0.001). There was no significant difference in ALP level between Yougui Pill group and normal group and sham operation group (P > 0.05). The level of ALP in the experimental group was significantly lower than that in other groups (P < 0.001). (2) Urinary Ca / Cr and urinary P / Cr in the model group were significantly higher than those in the normal group and sham operation group (P < 0.001). There was no significant difference in Ca / Cr and P / Cr between Yougui Pill group and normal group and sham operation group (P > 0.05). Ca / Cr and P / Cr in the experimental group were significantly lower than those in other groups (P < 0.05). (3) PINP and BMP-2 in the model group were lower than those in other groups, and the difference was statistically significant (P < 0.001). The concentrations of PINP and BMP-2 in the experimental group were higher than those in Yougui Pill group and model group (P < 0.05), but there was no significant difference with normal group and sham operation group (P > 0.05). (4) The bone mineral density of the model group was significantly lower than that of other groups (P < 0.001). The bone mineral density of the experimental group was significantly higher than that of the model group and Yougui Pill group (P > 0.05), but there was no significant difference with the normal group and sham operation group (P > 0.05). The bone strength of the model group was significantly lower than that of other groups (P < 0.001).. Conclusion The method of tonifying kidney and spleen, benefiting essence and filling marrow has the effect of anti-osteoporosis, and the mechanism may be related to adjusting the intestinal micro ecological environment, reducing the loss of bone minerals, up regulating the expression of BMP-2 in bone tissue and increasing the activity of osteoblasts.
