Abstract:Objective To investigate the apoptosis-inducing effect of ginsenoside CK on human osteosarcoma cell line MG-63 and its specific mechanism. Methods The experiment was divided into negative control group (Control group) and experimental group (CK drug group), MTT methods for detecting cell viability and proliferation; Apoptosis morphology detection, Hoechst nuclear staining and Annexin V/PI apoptosis assay were used to detect the apoptosis-inducing effect of CK on cells; Western blotting for Caspase-9, Bcl-2 and BAX expression. Results CK can significantly reduce the activity and survival rate of MG-63 in vitro(P<0.05); Apoptosis morphology detection, Hoechst nuclear staining and Annexin V/PI cell apoptosis assay demonstrated that CK can induce apoptosis of MG-63 (P<0.05); The expression of Caspase-9 and BAX in MG-63 cells was up-regulated and the expression of Bcl-2 was down-regulated after CK treatment. (P<0.05). Conclusion Ginsenoside CK has an apoptosis-inducing effect on human osteosarcoma cell line MG-63, and the mitochondrial apoptotic pathway with Caspase-9 as a key factor plays a decisive role in this apoptosis.
