Abstract:Objective To improve the instability of DBM activity due to donor quality by BMP quantitative. Methods DBM sample was obtained after soft tissue removal, degreasing, decalcifcation from the donor bone. The BMP-2 content was detected by Elisa method. Osteoinductivity was evaluated by implantation test in nude mice. The relationship between BMP-2 content and osteoinductive activity were observed. Results The BMP-2 content in different donor DBMs was quite different, the lowest is 5.85ng/g, and the highest is 71.83ng/g. There were signifcant differences on osteoinductivity in 4 weeks (P<0.05). Histological results showed that the bone formation area was increased with the increase of BMP-2 content. When the BMP-2 content was below 30 ng/g, bone formation area was <5%. When the BMP-2 content was more than 60 ng/g, the osteogenic area was >25%. Conclusion The content of BMP-2 was positively correlated with donor quality. Donor bone activity can be initially screened by Elisa.
